Nobody was injured in the attack, which shattered a large pane of glass and sent campaign workers diving for cover at the office near the Colorado state capitol building.
Police said they were not aware of any threats to the Obama campaign in Colorado.
The state is one of the closest-fought in the presidential election between Mr Obama and his challenger Mitt Romney. Both candidates have campaigned frequently in Colorado and faced off in the first presidential debate in Denver on Oct 3.
Det Raquel Lopez, a Denver police spokesman, said that officers were studying close circuit television footage to try to identify a vehicle from which the shot may have been fired.
"Right now, no one is in custody and investigators are looking at surveillance video to see if a vehicle can be identified," said Det Lopez said.
